Preparation and characterization of a tritiated derivative of CP-96.345-a nonpeptide antagonist of the substance P receptor-with a high specific radioactivity

A tritiated derivative of CP-96.345 with a specific activity of 84 Ci/mmole has been obtained. The radioactive compound has been characterized in comparison with the unlabeled analogue for its capacity for inhibiting the binding of the [{sup 125}I]iodinated derivative of substance P with brain membrane preparations from various sources. The capacity of [{sup 3}H]CP-96.345 for directly and specifically binding with membranes from the same sources has also been investigated by the method of radioligand analysis.
