Title: Oceanic heterotrophic dinoflagellates: distribution, abundance, and role as microzooplankton

Abstract

The primary objectives of this thesis were to determine the distribution and abundance of heterotrophic dinoflagellates across the Gulf Stream system off Cape Hatteras and to assess the potential grazing impact of these microheterotrophs in plankton communities. A list of species encountered in this study and their trophic status based on epifluorescence is presented, as well as observations on the presence of external or internal symbionts. The abundance of heterotrophic dinoflagellates across the Gulf Stream region off Cape Hatteras was determined from bimonthly net tow samples over a year and from whole water samples in March. Their average abundance was twice that of net ciliates in the net plankton and ten times that of ciliates in the nanoplankton. An isotope technique was developed to measure grazing rates of individual dinoflaggellates and other microzooplankton which cannot be separated in natural populations on the basis of size. /sup 3/H-thymidine and /sup 14/C-bicarbonate were used to label natural heterotrophic (bacteria and bacterivores) and autotrophic (phytoplankton and herbivores) food, respectively. Estimates of the grazing impact of heterotrophic kinoflagellates relative to other groups of heterotrophs on phytoplankton and bacteria were made by combining abundance data and clearance rates.   • The purpose of this study was to explore the role of serum protein binding and tissue distribution in the non-linear pharmacokinetics of warfarin in rats. The first phase of the research was an attempt to elucidate the causes of intersubject differences in serum protein binding of warfarin in rats. It was found that the distribution of S-warfarin between blood and liver, kidneys, muscle, or fatty tissue was non-linear. Based on the tissue distribution data obtained, a physiologically-based pharmacokinetic model was developed to describe the time course of S-warfarin concentrations in the serum and tissues of rats. The proposed model wasmore » able to display the dose-dependent pharmacokinetics of warfarin in rats. Namely a lower clearance and a smaller apparent volume of distribution with increasing dose, which appear to be due to the presence of capacity-limited, high-affinity binding sites for warfarin in various tissues. To determine if the binding of warfarin to the high-affinity binding sites in the liver of rats is reversible, concentrations of S-warfarin in the liver and serum of rats were monitored for a very long time after an intravenous injection of a 1 mg/kg dose. In another study in rats, non-radioactive warfarin was found to be able to displace tissue-bound C/sup 14/-warfarin which was administered about 200 hours before the i.v. injection of the non-radioactive warfarin, showing that the binding of warfarin to the high-affinity binding sites in the body is persistent and reversible.« less
  • Rates of bacterial production of particulate organic carbon in two hardwater Indiana lakes were studied. Primary production rates were calculated from rates of photosynthetic H/sup 14/CO/sub 3//sup -/ incorporation and bacterial (secondary) production from rates of /sup 3/H-(methyl)-thymidine incorporation by natural samples. The relationship of thymidine incorporation to rates of bacterial growth in diluted natural samples was used to calculate the conversion factor 2.2 x 10/sup 18/ cells produced (mole thymidine incorporated)/sup -1/. Bacteria in Little Crooked Lake were found to be growing at suboptimal temperatures throughout most of the water column, even during the summer months. Even rapidly growingmore » metalimnetic populations displayed no noticable adaptation to low environmental temperatures. This indicates that temperature could have limited the growth of bacteria in this lake throughout the period of thermal stratification. The extracellular release of photosynthetically fixed organic carbon by healthy phytoplankton was not found to be an important carbon source for planktonic bacteria. Slow carbon release mechanisms, such as algal decomposition, seem to have been more important.« less
  • In this dissertation the amplitudes of critically refracted (Pn) seismic head waves are compared with those generated by synthetic seismograms to estimate the velocity gradients in the uppermost oceanic mantle. By using laboratory measurements of compressional wave velocity as a function of pressure and temperature for various rock and mineral samples, predictions can be made of a critical temperature gradient for these samples. If the earth's gradient exceeds this value then the temperature effects will decrease seismic velocity with depth. It is shown in this work that the critical gradients for all samples of materials present in the upper mantlemore » (dunite, harzburgite, eclogite) are exceeded by the oceanic geotherm. Therefore velocity must decrease with depth if the earth is homogeneous and non-porous. A suite of amplitude-distance curves is built up for several different gradients. These curves are compared with real data from four experiments in the Pacific Ocean over seafloors of ages 2, 25, 40, and 65 million years. A number of petrologic models are discussed as explanations for the apparent positive gradient at the 65 my site. Most of the popular models are not adequate. One possibility is the presence of basaltic residuum in the upper mantle, decreasing with depth. Another is the possibility of slight serpentinization of the upper mantle with depth, requiring either native or meteoric water to be present in the upper mantle. The data alone cannot discern between these and/or other models as a solution to this problem.« less
