Laboratory apparatus for preparing and dispensing nitrogen-15-labeled anhydrous ammonia

An apparatus and a procedure are described for preparing liquid anhydrous NH/sub 3/ (LAA) of a desired /sup 15/N concentration and a cold-jacketed syringe for dispensing the LAA. Preparation of LAA was accomplished by reacting /sup 15/N-labeled (NH/sub 4/)/sub 2/SO/sub 4/ (30-50 atom % /sup 15/N) with a 2 M NaOH solution and allowing the resultant /sup 15/N-labeled NH/sub 3/ gas to mix with reagent grade anhydrous NH/sub 3/ gas dispensed from a lecture bottle (38.5- by 5.1-cm cylinder). The resulting gas mixture was condensed and drained into a collection vessel from which it could be withdrawn, measured, and dispensed with specially built cold-jacketed syringes. With the apparatus and procedure described here, 10 mL of LAA containing 0.7 to 2.0 atom % /sup 15/N can be prepared and applied quantitatively in amounts ranging from 25 (15.21 mg NH/sub 3/-N) to 500 ..mu..L (296.98 mg NH/sub 3/-N). A detailed description of how to construct and operate the /sup 15/N-labeled LAA preparation apparatus and dispensing syringes are discussed.
