Chromatographic characterization and biological behavior of technetium diphosphonate radiopharmaceuticals

This work describes the characterization of technetium-99m radiopharmaceuticals prepared with the ligand 2,3-dicarboxypropane-1,1 diphosphonate (DPD) and investigations into the biological behavior of other technetium diphosphonate radiopharmaceuticals. The disphosphonate ligands which were used in these studies include: methylene diphosphonate (MDP), 1,1-hydroxyethylidene diphosphonate (HEDP), dimethylaminomethylene diphosphonate (DMAD) and DPD. The sodium borohydride reduced technetium-99m dicarboxypropane diphosphonate (/sup 99m/Tc(NaBH/sub 4/)-DPD) radiopharmaceutical preparations were characterized and separated by anion exchange high performance liquid chromatography (HPLC). Several parameters important to the formulation of these /sup 99m/Tc-DPD radiopharmaceuticals were varied, including pH, presence or absence of atmospheric oxygen, concentration of technetium, and time post reaction.
