Nutrient-carcinogen interactions: the effect of oleate on the export of benzo(a)pyrene metabolites from the perfused rat liver

The effect of oleate on the production, accumulation and export of phenolic benzo(a)pyrene (BP) metabolites (free + glucuronides) was studied in perfused livers from ..beta..-naphthoflavone-treated rats. In the absence of bile salts, metabolites from BP (20 ..mu..M) were exported into the effluent perfusate at rates around 80 nmol/g/h and accumulated in the cell at rates greater than 200 nmol/g/h. Rates of export of free phenolic metabolites into perfusate were increased significantly 2- to 3-fold by oleate (500 ..mu..M) both in the presence and absence of the bile salt, taurocholrate (57 ..mu..M). With taurocholate, metabolites were exported into the bile at high rates (> 100 nmol/g/h). In the absence of bile salts, oleate did not influence export of metabolites into bile or accumulation in the liver. With taurocholate present, however, fatty acids decreased the rate of export of glucuronides in the effluent perfusate from 72 to 15 nmol/g/h and decreased accumulation in intracellular stores 4-fold. Metabolites increased slightly in the bile under these conditions. Under all conditions studied, overall rates of monooxygenation of BP were not affected by oleate. These data are consistent with the hypothesis that fatty acids displace non-covalently bound free phenols and glucuronides of BP from intracellular stores into the blood and bile in the presence of the natural, mild detergent action of bile salts where they may contribute to fatty acid-stimulated carcinogenesis.
