Tunicamycin inhibits ability of irradiated L. braziliensis promastigotes to protect against infection with virulent organisms in vivo
Intravenous immunization with irradiated promastigotes of L. braziliensis can protect BALB/c mice from the progressive, non-healing lesions caused by intradermal inoculation of virulent L. braziliensis. If mice are previously challenged with irradiated parasites via a subcutaneous route before intravenous innoculation no such protection is observed. They have examined the effect of inhibition of N-glycosylation of glycopeptides on the ability of irradiated promastigotes to produce these effects. At 1..mu..g/ml tunicamycin caused minimal (< 10%) suppression of /sup 3/H-leucine incorporation into parasite membranes, yet a 40%-60% inhibition of incorporation of /sup 3/H-mannose. Swainsonine, as expected, at 1-2..mu..M affected neither metabolic labelling procedure. Tunicamycin-treated promastigotes were unable to immunize mice for protection, but were apparently as effective as untreated organisms in blocking protection (by intravenous immunization) when given subcutaneously. These data are consistent with the notion that the protection inducing determinants are dependent upon N-glycosylation for their expression, unlike the determinants responsible for suppression of protection.
