Placental and lymphocyte markers of human exposure to polychlorinated biphenyls and polychlorinated dibenzofurans

Human exposure to toxic halogenated aromatics has occurred through accidental contamination of the food and the environment. One of the most severe episodes occurred in 1979, when rice oil contaminated with polychlorinated biphenyls (PCBs) and polychlorinated dibenzofurans (PCDFs) was consumed by approximately 2000 people in several counties of Taiwan. Total ingestion was estimated at 1-3 g PCBs and 0.5 mg PCDFs per individual. This unfortunate episode provided an opportunity to evaluate biochemical changes in a human population exposed to toxic halogenated aromatics. The objectives of their study were to evaluate placentas from exposed women for PCB and PCDF tissue concentrations, cytochrome P-450 isozymes, Ah receptor and epidermal growth factor (EGF) binding properties. Placentas were obtained approximately four years after the exposure had occurred. All women were non-smokers, were not taking any medications and had no recognized radiation or chemical exposure. The average age for both the exposed and non-exposed (women residing in the same counties) was 34 years. Placental homogenates and microsomes from exposed subjects showed marked elevation of aryl hydrocarbon hydroxylase (AHH) activities compared to control samples. Western blot analysis of placental microsomes prepared from exposed individuals revealed a protein which was immunochemically-related to rabbit lung cytochrome P-450 form 6, an isozyme induced by polycyclic aromatic hydrocarbons.
