Alveolar flooding and pulmonary endothelial extractions in tracer water, sodium ion and antipyrines in vivo in dogs
With the multiple indicator dilution technique they have examined in anesthetized, artificially ventilated, paralyzed dogs the extractions (E) and calculated permeability-surface area (PS) products for tracer water (THO), /sup 22/Na/sup +/, /sup 14/C-antipyrine (AP) and /sup 125/I-iodoantipyrine (IAP). After an initial control run, about 10 cm/sup 3/ x (kg body wt)/sup -1/ of an isotonic, buffered, protein-containing solution was rapidly introduced into the airway by gravity. One minute later, a second indicator-dilution was carried out. Mean values for E/sub Na/ and (PS)/sub Na/ did not differ significantly between the two successive runs. Average E/sub THO/ increased from about 0.54 to about 0.74. E/sub AP/ and PS/sub Na//E/sub THO/ fell from 0.95 and 1.15 to 0.85 and 1.00 respectively. The decrease of the extraction ratios could be interpreted indicating that a permeability barrier for AP and IAP had become evident. More likely, it results in major part from lower diffusion coefficients of AP and IAP than of THO in added alveolar fluid. The distribution of AP and IAP is not endothelial barrier limited but is not strictly flow (or volume) limited: it is determined at least in part by extravascular or tissue diffusion.
