Title: Studies of ethanol in the lateral hypothalamus of rats using in vivo microdialysis

Microperfusion of discrete rat brain areas using miniature dialysis probes has been shown to be useful for measuring extracellular levels of endogenous small molecules, especially monoamines and amino acids. We have adapted this technique to monitor brain levels of ethanol following a single IP dose. Male Sprague-Dawley rats underwent surgery for stereotaxic placement of microdialysis guide cannulae in left lateral hypothalamus and catheterization of left jugular vein. The hypothalamus was perfused with artificial CSF at a rate of 1.9 uL/min. Following a 1 hr period of baseline sampling, rats were injected IP with ({sup 14}C)-ethanol (20 uCi) at a dose of 0.8 or 2.4 g/kg (20% w/v in saline) and collection of perfusates was continued at 10 min intervals. Plasma was prepared from blood drawn through the catheter every 10 min and the radioactivity content of perfusates and plasma determine by scintillation counting. Following injection of the lower dose, ethanol was detected in the brain by 10 min, peaked at 20-30 min and then declined slowly. After the higher dose, ethanol was also detectable by 10 min, peaked at 20 min and remained close to peak levels for over 1 hr before declining slowly. These results indicate that microdialysis is a useful technique for monitoring brain levels over time.
