High density lipoprotein (HDL) cholesteryl ester and protein metabolism in copper-deficient hypercholesterolemic rats

Weaning male Sprague-Dawley rats were divided into two dietary treatments; copper-deficient (CD) and control (0.8 and 6.5 {mu}g Cu/g diet, respectively). After 6 weeks, animals from each group were injected with labeled HDL previously isolated from rats subjected to the same dietary treatments. Cholesteryl esters were labeled by incorporating ({sup 3}H)cholesteryl linoleyl ether (CLE) into the particle core, which served as a non-degraded marker of tissue uptake. The protein moiety was radiolabeled with {sup 125}I. Kinetic analysis of doubly labeled HDL indicated that ({sup 3}H)CLE was cleared from the plasma faster than {sup 125}I-protein in CD and control rats. Because CD animals exhibited significant increases in the plasma pool size of HDL components, absolute mass clearance and subsequent tissue uptake were determined. When expressed as {mu}g/hr per organ per 100 g body weight, the liver was clearly the most important organ for ({sup 3}H) CLE uptake in both treatment groups; moreover, ({sup 3}H)CLE uptake in the liver of CD rats were 2.1-fold higher than controls. The skin and muscle were the major sites of {sup 125}I-protein uptake in both treatment groups, although uptake was greater in CD rats. That HDL cholesteryl ester transport to the liver was increased in copper deficiency, and in view of previous studies demonstrating no increase in cholesterol excretion, these data suggest that net flux of HDL cholesterol may be increased and that HDL production by the liver may also be increased in this hypercholesterolemic model.
