Studies in iodine metabolism: Progress review, 1954-1964

We have developed an I-131 labeled diet and studied the iodine equilibrium in experimental animals. These studies led to major investigation into thyroxine metabolism in which we used chemical iodine analyses and I-131 methods, to study metabolism of ingested thyroid glands and administered I-131 labeled thyroxine and its analogues. One important result was the discovery of an inhibitory effect of propylthiouracil upon thyroxine metabolism. These studies on iodine metabolism led to the suggestion that there might be a pool of iodine in thyroid glands which turned over very slowly compared to the major metabolic pools and which is very slowly hydrolyzed in vivo. In the past year we have clearly demonstrated such a system in hyperplastic thyroids. We have shown that most of the thyroid iodine will leave the gland with a biological half-life of 10 hours but 1 to 20% will have a half-life that is 10% as long as the entire normal life span of the rat.
