Antisera reactive with the N-formyl peptide receptor of rabbit neutrophils

Of central importance in the biology of the neutrophil is the cell's capacity to migrate to sites of tissue damage or bacterial infection. This migration is guided by the interaction of chemotactic peptides such as f-Met-Leu-Phe with specific receptors on the cell surface. Since extensive purification of these receptors by biochemical methods has been largely unsuccessful the authors have begun to develop antisera reactive with this receptor. It has been possible to raise both polyclonal antisera (in guinea pigs) and mouse monoclonal antisera reactive with the rabbit receptor for f-Met-Leu-Phe. Antisera were obtained after immunization with receptor enriched preparations. These antisera were screened by ELISA and FACS analysis for reactivity with neutrophil membranes and for anti-receptor activity by 1) their ability to precipitate CHAPS-soluble receptor previously complexed with f-Met-Leu-(/sup 3/H)Phe and 2) receptor covalently cross-linked to the radioiodinated chemotactic peptide, f-Nle-Leu-Phe-Nle-(/sup 125/I)Tyr-Lys. Currently they are attempting immunoaffinity purification of the neutrophil receptor for chemotactic formylated peptides using these different groups of antisera.
