Proliferation of T cell clones requires signals in addition to the pertubation of the T cell receptor - CD/sup 3/ complex

Antigen-specific activation of T lymphocytes is mediated through pertubation of the T cell receptor - CD3 complex which initiates a characteristic cascade of intracellular events including induction of IL-2 secretion, expression of interleukin-2 receptor and finally, T cell proliferation. It has been shown that the antigen-receptor interaction can be mimicked by binding of anti-CD3 antibodies in the presence of phorbol-esters. To study the sequellae of events after antigenic stimulation, they generated a panel of alloreactive human T cell clones all of which were functionally characterized by a strong proliferative response when co-cultured with alloantigen. Based on their proliferative response to monoclonal antibodies against CD-3 plus PMA or PMA plus PHA, they could clearly divide the clones into two groups. One group had a normal proliferative response to the mitogenic signal of monoclonal anti-CD3 antibodies. The other group failed to proliferative when stimulated by antibodies or mitogens. The lack of proliferation in the latter clones was not correlated with an abnormal regulation of intracellular calcium flux or the expression of the CD-3 molecule on the cell surface. In fact, mitogenic stimulation could initiate IL-2 secretion, as well as the expression of IL-2 receptors but not proliferation in these clones. Their data provide evidence that activation by antibody to CD-3 and PMA is not sufficient to induce proliferation in all T cells. Additional signal(s) provided by antigenic stimulation are required to induce T cell growth.
