Preparation of C-11 labeled dimethylsulfoxide (DMSO)

Dimethylsulfoxide (DMSO) is used as a carrier for percutanous resorption of various pharmaceuticals. Besides the evaluation of its carrier characteristics, C-11 labeled DMSO may allow the determination of peripheral blood flow by measurement of the kinetics of DMSO skin penetration, its disappearance at the site of application and its appearance in blood. Routinely produced n.c.a. (C-11)-methyliodide (ca. 20 GBq(540 mCi)) is collected in 100 ..mu..l acetone. The precursor is mixed with 10 ..mu..Mol sodium-sulfide in 100 ..mu..l water and reacted for 5 min. at 60/sup 0/C. Thereafter 20 ..mu..Mol methyliodide carrier in 50 ..mu..l acetone are added and the reaction is continued for 5 min. The rection generates highly volatile dimethylsulfide which is carried with a stream of nitrogen into a small vessel containing 100 ..mu..Mol hydrogenperoxide in 200 ..mu..l acetone. The oxydation to DMSO proceeds at room temperature and is complete within additional 5 min. The acetone is then evaporated and the aqueous residue is purified by loading it onto a C-18 reversed phase cartridge and elution with 10 ml of physiol. NaCl solution.
