Effects of endogenous picolinic acid on zinc absorption

Male Sprague Dawley rats (250 g) were fed 20% egg white diets containing either 5 ppm or 20 ppm Zn for a total of 4 weeks. After 2 weeks of dietary regimen, each dietary group was divided into three treatment groups receiving continuous intravenous infusion of either 0.9% NaCl (CON) or picolinic acid (PA) solutions containing 200 mg PA/ml (PA 200) or 50 mg PA/ml (PA 50) from subcutaneously implanted osmotic pumps. The doses of PA delivered were 100 ..mu..g/hr (PA 200) and 25 ..mu..g/hr (PA 50). /sup 65/Zn was used to measure true Zn absorption by isotope dilution. Both levels of PA increased dietary Zn absorption from 66 +/- 7% (CON) to 73 +/- 4% (PA 200) and 76 +/- 5% (PA 50) (values are means +/- SEM; CON vs PA 200, NS; CON vs PA 50, p = 0.01). Serum Zn (..mu..g/ml) increased from 1.27 +/- 0.08 (CON) to 1.48 +/- 0.06 (PA 200) and 1.54 +/- 0.04 (PA 50) (CON vs PA 200, p = 0.002; CON vs PA 50, p = 0.0001), and serum specific activity of /sup 65/Zn (nCi/..mu..g Zn) was decreased from 0.64 +/- 0.05 (CON) to 0.52 +/- 0.06 (PA 200) and 0.58 +/- .04 (PA 50) (CON vs PA 200, p = 0.009; CON vs PA 50, NS). Tibia Zn (..mu..g/g dry wt) was increased from 109.2 +/- 5.4 (CON) to 119.4 +/- 3.4 (PA 200) and 117.1 +/- 4.9 (PA 50) (CON vs PA 200, p = 0.04; CON vs PA 50, NS). The effects of PA were not influenced by dietary Zn levels. Also, neither level of PA affected either intestinal or urinary Zn excretion. These results suggest that endogenous PA influences the absorption and utilization of dietary Zn.
