Analysis for polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF) in human adipose tissue: method evaluation study. Final report

The report focuses on the evaluation of an HRGC/MS analytical method for determination of 2,3,7,8-substituted polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF) in human adipose tissue. This method will be used for analysis of samples from EPA's National Human Adipose Tissue Survey (NHATS) as part of a collaborative effort between EPA's Office of Toxic Substances and the Veterans Administration. The method was evaluated using aliquots of a bulk lipid matrix that was extracted from human adipose tissue. The results of the replicate analysis of spiked and unspiked homogenized human adipose tissue matrix demonstrate that the analytical method produces accurate and precise data for 17 specific 2,3,7,8-substituted PCDD and PCDF (tetra- through octachlorohomologs) congeners. The endogenous or background levels of the PCDD and PCDF congeners in the homogenized adipose lipid matrix were estimated through regression analyses of measured versus spiked concentrations for each compound. This unspiked matrix will be used as a control sample with each batch of samples analyzed.
