Ontogeny and site of intestinal lipid and lipoprotein synthesis

The cellular mechanisms leading to the formation of intestinal lipoproteins remain obscure. Our objective was to determine the ontogeny of intestinal lipoprotein synthesis in the jejunum and ileum. Explants of jejunum and ileum from suckling (5 d) and weanling (23 d) were cultured in the presence of {sup 14}C-oleic acid to examine lipid synthesis (DPM/mg protein). Incorporation of oleate by jejunum and ileum was significantly increased for the weanling compared to suckling rats. Triglyceride synthesis followed the same pattern, but only for the jejunum, whereas the latter group's jejunal cultures produce more VLDL-TG. The incorporation of {sup 14}C-oleate as a function of site revealed increased uptake by ileum compared to jejunum by 24% and 36% for weanling and suckling groups respectively. Furthermore, at the suckling period synthesis of TG, CM-TG and VLDL-TG was similar for jejunum and ileum explants, whereas it was elevated at the jejunum compared to the ileum for the weaning period. Our findings show important changes that occur in lipid and lipoprotein synthesis during development, and that differences exist between ileum and jejunum. The modification may be influenced by alterations known to occur during this period.
