Incorporation of dinitrophenyl-modified proteins within the 30S subunit of E. coli ribosomes

Proteins were modified to varying extents (0.5 equivalent or more) with 2,4-(3,5-/sup 3/H)-dinitrofluorobenzene (DNFB) and separated from unmodified protein by RP-HPLC using dual wavelength monitoring at 214 and 360 nm. DNP-modified proteins were next reconstituted into 30S subunits as described by Held. Subunits were purified by sucrose gradient sedimentation and size exclusion-HPLC. The presence of DNP-modified protein was detected by radioactivity co-sedimenting with the 30S particle. RP-HPLC analysis of proteins extracted from 30S subunits containing modified proteins show a full complement of proteins.
