Title: Investigation of the biotransformation of promutagens by the Ames test. Communication II. Degree of mutagenic action of nitrosomorpholine, diethylnitrosamine, and cyclophosphane, using various subfractions of a rat liver homogenate

The work presents data on the participation of the components of the microsomal and cytosol subfractions of the S-9 fraction of rat liver homogenates in processes leading to the formation of active metabolites of promutagens nitrosomorpholine (NM), diethylnitrosamine (DENA), and cyclophosphane (CP) and their detoxification as a result of the reaction with glutathione (G-SH), added to the system. It was established that the metabolic activation of NM is associated only with the microsome subfractions, whereas the reactions of the first phase of CP and DENA metabolism occur when both microsomal and cytosol subfractions are added. A decrease in the effect of all the promutagens studied under the influence of G-SH was added when the microsomal and cytosol subfractions of the fraction S-9 were introduced into the activating mixture. Various dependences of the level of metabolic activation and degree of reduction of the mutagenic effect from the addition of G-SH on the protein content in the microsomal and cytosol subfractions were detected for the investigated promutagens.
