Kinetic characterization of /sup 13/NH/sub 3/ and (/sup 13/N)glutamine metabolism in rabbit heart

The isolated arterially perfused rabbit interventricular septum was used to study the effects of methionine sulfoximine (MSO) on /sup 13/N-labeled ammonia (/sup 13/NH/sub 3/) and /sup 13/N-labeled glutamine ((/sup 13/N)) metabolism. Tissue time-activity curves were generated from bolus injections of /sup 13/NH/sub 3/ or (/sup 13/N)gln delivered interarterially during control conditions and in the presence of MSO. The fraction of /sup 13/N label retained in the tissue (EF(3)) was significantly less after a bolus of (/sup 13/N)gln than /sup 13/NH/sub 3/. MSO significantly decreased the EF(3) after a bolus of /sup 13/NH/sub 3/ or (/sup 13/N)gln. Chemical analyses using high-pressure liquid chromatography on tissue and effluent samples obtained after bolus injections of /sup 13/NH/sub 3/ and (/sup 13/N)gln with or without MSO present showed the following: 1) after a bolus of /sup 13/NH/sub 3/ or (/sup 13/N)gln the tissue /sup 13/N label is predominantly in (/sup 13/N)gln at 6 min after injection, and 2) MSO significantly decreases the synthesis of (/sup 13/N)gln after an /sup 13/NH/sub 3/ bolus. Thus it would appear that the glutamine synthetase reaction is primarily responsible for the retention of the /sup 13/N label from /sup 13/NH/sub 3/ as (/sup 13/N)gln in rabbit myocardium.
