Antioxidant response of eastern white pine, Pinus strobus, to ozone exposure

The authors have characterized components of antioxidant metabolism in two clones of eastern white pine that were selected as tolerant or sensitive to environmental stress. In order to identify how air pollutants affect these phenotypes, clonal plants were fumigated with ozone over 4 days during the 16 h light period. Ozone concentrations were increased by increments of 20 ppb from the initial concentration of 25 ppb to 150 ppb at mid-day (1,200-1,400 h) and returned to ambient by the dark period (2,200 h). Glutathione reductase (GR) activity and soluble protein initially decreased in response to O{sub 3} exposure. GR protein was quantified with antibody to the major form of GR in pine. Compared to controls, GR protein was 33 times greater in the sensitive clone and 133 times greater in the tolerant clone after 4 days treatment. Superoxide dismutase (SOD) activity increased 60% only in the more sensitive clone. Data suggest that sensitivity to oxidative stress in white pine may relate to uncoordinated expression of enzyme activities responsible for antioxidant metabolism. Metabolic flux through the pathway may be limited by GR activity.
