Preparation and characterization of nitrogen-sulfur donor ligands and their technetium complexes

Technetium complexes of bidentate aminoethanethiol and tetradentate bis-aminoethanethiol ligands were studied to determine the charge, stoichiometry, lipid solubility, stability, and structure of the complexes, technetium-99m and technetium-99 were used in a complementary fashion to obtain the information necessary to determine the suitability of using these ligands for incorporation into biologically interesting compounds for radiotracer development. Bidentate aminoethanethiol ligands form bis complexes with technetium of the general form: TcOXL/sub 2/. These complexes is stable in solution; however, they rapidly exchange with 1,2-ethanedithiol to form the bis(ethanedithiolato) complex. Tetradentate bis-aminoethanethiol ligands form 1:1 complexes with technetium of the general form: TcOXL. These complexes are also stable in solution and exchange very slowly with 1,2-ethanedithiol. The synthesis of the ligands and complexes are described and the implications of the results obtained on radiotracer development are discussed.
