Title: Absence of effects of phenobarbital pretreatment or multiple benzene exposures on in vivo biotransformation of benzene in mice

The authors have previously reported a concentration-dependent shift in the sulfation and glucuronidation of phenol for mice exposed via inhalation to constant benzene concentrations ranging from 35-600 ppm for 6 hours. Pretreatment of the mice with phenobarbital did not alter the blood levels of benzene, phenol, phenylglucuronide or phenylsulfate when compared to saline injected animals. Mice were also exposed to various benzene levels for 6 hr/day x 5 days. Once again, no significant difference in the blood concentration of phenol or its conjugates was found when compared to mice exposed for only 6 hours. In all cases, phenol and phenylglucuronide blood concentrations increased linearly through 600 ppm when compared to blood concentrations of benzene while sulfation was apparently saturated above 200 ppm. These data suggest that blood concentrations of phenol and its conjugates are not changed by either phenobarbital pretreatment or multiple benzene exposures. Toxicity in the 5 day exposures was measured as a decrease in total white blood cells (WBC) in the peripheral blood compared to sham exposed controls. Total WBC decreased steadily from 3 ppm benzene up to 200 ppm. No further decrease in WBC was seen above 200 ppm.
