The kinetics of 2,2 prime -dichlorodiethyl sulfide (sulfur mustard, HD) dependent cell death

Human lymphocytes exposed to HD demonstrate a concentration dependent decrease in cellular viability. In order to design assays to determine the efficacy of candidate antidotes to prevent HD cytotoxicity, it was necessary to study both time and concentration-effects of HD exposure. The kinetics of HD initiated death was studied by determining propidium iodide exclusion by human lymphocytes over a concentration range from 10{sup {minus}6} to 10{sup {minus}3}M HD. An automated system was developed using 96 well plates in a flow cytometer. This system enables the simultaneous study of both concentration- and time-dependant effects of HD on cytotoxicity. HD caused a non-linear multiphasic decrease in cellular viability with time. During the first 4 hours, there was no decrease in lymphocyte viability at any exposure level. After this latent period, the rate of cellular death was proportional to the concentration of HD during the next 10 hours. During 14-20 hours post exposure period, there was a leveling off of the rate of cell death at the higher concentrations of HD. Thus, it was determined that an incubation period of 20 hours post-exposure to 3 {times} 10{sup {minus}3} M HD would be employed for investigations of the efficacy of possible antidotes to HD induced cytotoxicity.
