INFLUENCE OF COBALT-60 RADIATION ON HEMOGLOBIN FORMATION IN THE CHICK EMBRYO

Fertile, 96-hr hen eggs (White Leghorn and New Hampshire breeds) were irradiated with 100-800 r from a Co/sup 60/ source. Hemoglobin was obtained from the chicks at 1, 4, 7, and 18 days after hatching and fractionated by starch gel electrophoresis. Five hemoglobin fractions, designated I-V, were isolated from nonirradiated controls; no differences were found in relation to breed, but the electrophoretic patterns varied according to the age of the chicks. Irradiation with 500 or 700 r produced a new, rapidly migrating hemoglohin component (fraction IV), whose frequency of appearance increased proportionally with the radiation dose. With some doses the concentrations of components II, III, and IV diminished, possibly due to greater ease of oxidation of hemoglobin to methemoglobin in irradiated cases. Hematocrit was decreased by irradiation, to as low as one-third normal. The LD/sub 50/ of irradiation in these experiments was found to he 700-800 r, and with doses exceeding 600 r the chicks exhibited central nervous system disorders and deformities, particularly of the extremities.
