SPECIFIC RADIOPROTECTIVE EFFECT OF THE MITOCHONDRIAL MATRIX
>Results of experiments on radiation effects on glutamic dehydrogenase without and within the intact mitochondrion are reported. When the intact mitochondrial suspensions were irradiated, no change in the total glutamic dehydrogenase activity could be found with doses up to 133,000 r and no significant activity was found in the supernatants from centrifuged irradiated and nonirradiated mitochondria. A purified, crystalline preparation of beef liver glutamic dehydrogenase was added to a suspension of intact mitochondria and the mixture irradiated. The activity of the free added emzyme was markedly diminished while that of the mitochondrial enzyme was unchanged. However, the presence of the mitochondria did afford some protection to the added enzyme. The results demonstrate that glutamic dehydrogenase contained within the mitochondrial matrix is remarkably well protected from the destructive effects of ionizing radiation. (P.C.H.)
