RADIOBIOCHEMICAL STUDIES ON DNA (in French)

The nature of the products formed when the pyrimidic bases in oxygenated aqueous solution are exposed to x or gamma radiation was determined. The reaction starts by peroxidation. The peroxide of thymine is stable; that of cytosine is very unstable and leads quickly to the terminal products 5- hydroxycytosine and isobarbituric acid. That of uracile is only slightly stable and rapidly yields glycol. Peroxidation in oxygenated media gives a high yield. In the absence of oxygen, the reactions are different. Pneumococcus-transforming DNA is inactivated more rapidly by x rays than by the 2537 uv radiation. The ratio of the quantum yields of the ionization and of this photon is equal to 5200. The DNA double chain fiber absorbs 13% of the 2537 photons which it receives. The DNA of a B/r bacteria was labeled with P/sup 32/, with C/sup 14/ in the 2 position on the thymine, and with H/sup 3/ on the CH/sub 3/ of the thymine. The lethality caused by this labeling was studied in these three cases at -196 and 0 deg C. The H/sup 3/ lethality is exponential. It is produced principally by beta rays. The C/sup 14/ lethality is exponential and is produced by decay. Its yield at -196 deg C is 1/75. P/sup 32/ lethality follows a sigmoid curve. The yield is 1/38 at -196 deg C and 1/11 at o deg C.
