Effects of ‘Visible’ Light on 5-Bromouracil-labelled DNA

Results are reported from a study of the effect of visible light on the incorporation of 5-bromodesoxyuridine, labeled with C/sup 14/, into the desoxyribonucleic acid (DNA) of intact cells of Bacillus subtilis, a thyminine- requiring strain of Escherichia coli, or protein- and RNA-free DNA isolated from genetically transforming strains of B. subtilis. The photochemical changes produced by a 36 hr illumination from a fluorescent lamp of labeled DNA were followed by determination of the chromatographic behavior of the C/sup 14/ label after 14 hr at 25 deg C or 1 hr at 95 deg C of 70% perchloric acid hydrolysis of the non-illuminated and illuminated DNA, and by measuring the effect of illumination on the buoyant density of labeled DNA. Similar experiments using longwave ultraviolet light gave essentially identical results, indicating that the effects of visible light are probably attributable to its ultraviolet components. Reaction mechanism involved in the dehalogenation during irradiation of labeled DNA was investigated by density measurements on the halogenated DNA before and after illumination.
