NUCLEAR PROTEIN SYNTHESIS IN VICIA FABA

A technique for the rapid isolation of cytoplasmic-free preparations of Vicia faba nuclei was developed. By using this process and autoradiography, differences in rate and pattern of nuclear incorporation of tritiated lysine, arginine, and tryptophan in the meristematic cells were studied. Lysine is incorporated rapidiy into the nucleus, and concentration is particularly evident in the nucleolus and perinucleolar chromatin. Arginine incorporation is somewhat retarded. Long exposures of autoradiographs are necessary to show tryptophan label. Lysine incorporation first becomes evident in the perinucleolar chromatin. The nucleoli become labeled, although the product accumulated is quite mobile and is readily washed out of the nucleoli in the presence of nonradioactive lysine. Chromatin label, on the other hand, may still be found at the end of 48 hr, although the amount of label is decreased. (P.C.H.)
