The Production of Chromatid Aberrations by 5-Fluorodeoxyuridine Alone and in Combination with X-Rays and 8-Ethoxycaffeine
In Vicia faba lateral root-tips cell divisions containing chromosomal aberrations began to appear approximates 3 hr after immersion of the roots in solutions containing FUDR in concentrations of 0.5 mu M or higher. The aberrations consisted mainiy of gaps and chromatid breaks. Aberrations involving rejoining (isochromatid breaks with sister union, exchanges) were extremely rare. FUDR aiso had a strong inhibitory effect on mitosis. Fifteen to 30% of the cells dividing 24-6 hr after a 1-hr treatment with 1 mu M FUDR contained chromatid aberrations of the usual type. The effects of FUDR increased with temperature and were stronger at low than at high pH. When x irradiation was combined with FUDR treatment, there was a marked increase in the number of gaps, chromatid breaks, and metaphases with extreme fragmentation in comparison with the x-ray control. When EOC was combined with FUDR, the effect was smaller than the sum of the effect of each alone. FUDR did not affect the frequencies of EOCinduced exchanges and isochromatid breaks with sister union. The effect of FUDR, on the other hand, was markedly reduced by EOC. (TCO)
