MUTAGENIC ACTION OF SOME RADIOMIMETIC AGENTS ON T2 COLIPHAGE (in German)
The mutagenic action of various cytostatic agents used in cancer therapy were assayed by comparing their effect on the mutation rate of T2h/sup +/ phage with the spontaneous rate. Myleran increased the rate 10 fold, and a lesser degree of mutagenicity was found with the following drugs (in decreasing order of effectiveness): A139 (2,5-bis-methoxyethoxy3,6-bis -ethyleneiminobenzoquinone), Mitarson, Endoxan, N-oxide mustard, N-methyl mustard, and N mustard. Their lethal doses in the phage and effect on Escherichia coli B generation time were also determined. The mutagenic action of the N mustards was correlated with the release of chloride ion. Mutagenic action of the agents tested was most pronounced at concentrations approximates 10/sup -5/M; at higher concentrations cytotoxic effects predominated over mutagenic ones. Demonstration of mutagenic activity of N mustards was carried out in phosphate buffer; it was confirmed that this activity is readily lost in bicarbonate buffer. (H.H.D.)
