The proposed explanation of the slowing down of glucose absorption in x- irradiated guinea pigs involves two factors active in normal animals: increasing glycemia and progressive dilution of ingested glucose in the stomach. To verify this hynothesis, guinea pigs were irradiated with unfiltered 250-kv x rays at a dose of 500 r. After 14 days they were fed glucose; the amourt of glucose absorbed/unit time was found to be a linear function, rather than the logarithmic one for normal animals. Glycemia/untt time was an exponential function for both normal and irradiated animals, with max value occurring at 70 min. In irradiated animals the max was 400 mg% as against 220 mg% for the control group. Concentrations of glucose in the stomach decreased from 140 mg/ml to an average of 56.2 plus or minus 12 mg/ml in control and 60 plus or minus 10 mg/ml in irradiated animals, in the first dilution period. A second stage of dilution occurs at the end of the first hour in normal animals and reduces the original 700 mg of glucose in the stomach at a rate exponential to the amount of glucose remaining. In irradiated animals this phase does not begin until near the experiment's end (the 3rd postabsorption hour). Hence the essential change in absorption after irradiation is that irradiated animals show decreasing volumes and stable concentrations of glucose, It was concluded that the slowing down of absorption at the end of the first postirradiation hour was not due, as once thought, to temporary inhibition of gastric drainage by glucose present in the stomach, but rather due to increased glycemia and decreased concentration. Intestinal absorption was found to be two phased, as are many other radioinduced functions. Delay in glucose metabolism correlates with the disturbance in digestive absorption of glucose in irradiated guinea pigs. (BBB)
