Methods for the analysis of human chromosome aberrations

Some of the information available from ionizing radiation studies (on which more information is available than on other mutagens) is presented and techniques for this work are described. In addition, some of the difficulties and pitfalls in the scoring of chromosome aberrations and in their utilization for biological dosimetry are described. Certain chemical mutagens can induce aberrations in human cells, and many of the technological problems encountered when chromosome and chromatid aberrations are used for estimating radiation dose are also relevant to the study of the chromosome-damaging effect of other mutagens. The study of a single individual, several individuals, or a population exposed to mutagenic agents is not a simple task. In the case of a single individual or several individuals, it is necessary to obtain good chromosome preparations and to score a minimum of 200 cells per person and a similar number of cells from matched controls. Such a study requires considerable time and expertise. A further aim of this manual is to describe standard methods for obtaining good chromosome preparations from human material and a system of scoring and data recording that is considered by an international group to be the best available at present. The use of this system would make it easier to compare data on chromosome aberration frequencies from different laboratories. (auth)
