Observations were conducted on 45 cases of leukemia including acute leukemia (AL) 33 cases (acute myelogeneous leukemia (AML) 24 cases, acute lymphatic leukemia (ALL) 9 cases, and chronic myelogeneous leukemia (AML) 12 cases) in order to study the factors that characterize the onset mechanism of leukemia. lncrease in monocytes and basophils in one to two years prior to onset of leukemia was observed in AL cases and was particularly remarkable in AML cases. In CML cases, the counts of white blood cells, neutrophils and basophils were all considerably high compared with the contrast group. Changes in basophils and staff neutrophils became gradually distinctive in three years prior to onset, the count being five to six times larger in one year before onset of leukemia. A gradual increase in Downey type and abnormal lymphocytes was observed beginning six years before onset in both ALL and CML cases. Increase in staff neutrophils two years prior to onset is considered to indicate the overproduction or accelersted tarn-over of granulocytes. One of the findings of interest was the increase in abnormal and atypical lynnphocytes in five to six years prior to onset. This can be interpreted that in preleukemic stage lymphatic tissue is destroyed by viras and other external antigens or stimulated by autoantigens produced by tissue exposed to radiation. It can also be considered as an immunological reaction of lymphoendothelial system attempting to exclude mutant clone induced by radiation. Close association of such mechanisms with the onset of leukemia is assumed considering the fact that latent period of two to five years of radiation-induced leukemia corresponds to the appearance of lymphocytes of these types.
