Analysis of a gluconeogenetic $sup 14$C and $sup 3$H tracer study. II (in Japanese)

ABS>Gluconeogenesis was analyzed using /sup 14/C- and /sup 3/H-labeled pyruvate, lactate, glycerol, and acetate. By the intravenous administration of 40 to 100 mu C of /sup 14/C and 200 to 300 mu C of /sup 3/H to diabetics and si mple obesities, the total /sup 14/CO/sub 2/ in the expired air and /sup 14/C and /sup 3/H in the blood glucose were measured. The /sup 14/CO/sub 2/ level in simple obesities and mild diabetics fell to about 50% of that of normal individuals and further in severe diabetics, showing a marked disturbance in glucose oxidation. When gluconeogenesis was observed using pyruvate-2-/sup 14/C, the incorporation of /sup 14/C into glucose was twice higher in mild diabetics and further higher in severe diabetics than in normal individuals, showing a reverse correlation to the decrease of breath-/sup 14/CO/sub 2/. Control of glycometabolism with insulin resulted in a remarkable decrease in the incorporation of /sup 14/C into glucose. Although the rate of incorporation of / sup 14/C in glycerol, an intermediary substance in fatty acid metabolism, into glucose was nearly the same as that of pyruvate, the rate of incorporation of tics. The results of the study of Shreeve, coworker, who analyzed in which position of the carbon of glucose the /sup 14/C came, were presented. Gluconeogcnesis is augmented through dicarboxylic shuttle and the rate of entry into the gluconeogenetic route tl::ough the TCA cycle was very low. The incorporation of /sup 3/H into glucose was similarly increased in diabetics to that of /sup 14/C, indicating the augmentation of gluconeogenesis. (JA)
