Title: THE GENETIC AND PHENOTYPIC CHARACTERISTICS OF RADIATION-INDUCED MUTATIONS IN MICE

Data ars presented on the genetic and phenotypic characteristics of 174 mutations at seven specific loci recovered after irradiation of mouse spermatogonia with three doses of acute x rays. There are several lines of evidence that suggest that mutations induced in spermatogonia are point mutations or very small deficiencies. The most cogent among these is the absence of simultaneous mutation of the closely linked d- and se-loci. Deficiencies involving the d- and se- loci occur ipontaneously and can be induced in postspermatogonisl stsges. Since they have been shown to be frequently capable of passsge through meiosis and of transmission, fallure to recover them from irradiated spermatogonia is evidence that they are not induced. Marked variation in mutation rates was found among the seven loci. In view of the fact that mutations recovered from irradiated spermatogonia appear to be mostly point mutations, this variation may iadicate intrinsic propenties of the loci or the closely adjacent regions. The specific locus method has detected a wide range of alleles, from those only slightly different from wild type, through apparent repeats of the test allele, to mutations lethal in homozygous coadition. This last class is the most frequent. Since deleterious effects of many of the individual lethals in heterozygous condition have been detected, however, and since unidentified mutations elsewhere in the genome have statistically demoastrable deleterious effects, it seems likely that heterozygous effects will add up to a greater total genetic damage in the population than the effects of homozygotes.
