INHIBITION OF RNA POLYMERASE BY 6-AZAURIDINE TRIPHOSPHATE

ABS>Escherichia coli RNA polymerase was prepared, and the synthetic reaction was assayed by the method of Chamberlin and Berg. Phosphorus-32-labeled uridine triphosphate (UTP) was used as the proximally labeled nucleotide. The pyrophosphate-nucleoside triphosphate exchange reaction dependent on crab- daT(deoxyadenylate and thymidylate) (or daT synthesized by Kornberg's polymerase) was maximal when UTP alone was included in the reaction. Labeled pyrophosphate- UTP exchange was significantly inhibited by aza-UTP. (H.M.G.)
