Effects of cadmium on human trophoblastic cells in vitro

This study was conducted to evaluate the bioaccumulation of cadmium in human trophoblastic cells in vitro at various stages of trophoblastic differentiation. Human placentae were obtained from selective Cesarian sections. Villous tissue was minced and enzymatically dispersed, and cytotrophoblastic cells were isolated on a Percoll gradient. Cytotrophoblastic cells were cultured in DMEM supplemented with 10% FBS at 37{degrees}C in a humidified atmosphere at 10% CO{sub 2} in air. Cells were cultured for varying time periods of 72 to 92 hours to permit various degrees of cell differentiation before adding 2 {mu}M cadmium in the form of cadmium chloride. Cells were harvested after 24 hour exposure to cadmium and were analyzed for cadmium content by atomic absorption spectrometry and for protein concentration. Results demonstrated that there was cadmium accumulation in the trophoblastic cells cultured for 72 hours (0.63ng Cd/{mu}g protein) while no cadmium was measured in the cells of the 92 hour culture. This suggests that cadmium accumulation is greater in the cytotrophoblastic cells than the syncytiotrophoblastic cells which indicates a potential toxic effect on placental development in vivo.
