Assessment of the bioaccumulative properties of a fluorinated anionic surfactant, ammonium perfluorooctanoate, using Pimephales promelas

Juvenile fathead minnows (Pimephales promelas) were exposed under static conditions to 25 mg/L of a fluorinated anionic surfactant, ammonium perfluorooctanoate (PFO), for 13 days. Following the uptake portion of the experiment, fish were transferred to clean water for a 15 day deputation phase. Whole fish and test solution samples taken during the experiment were combusted in a modified organic halide analyzer. Fluoride released from the PFO molecule during combustion was collected in a solution of TISAB and water. Free fluoride in the collection fluid was measured with an ion-selective electrode, and the concentration of PFO in whole fish tissue and test solution was determined by calculation. The concentration of PFO in test solutions from the static system remained constant at 25 mg/L and < 1.0 mg/L during the uptake and deputation phases, respectively. After 1 3 days of exposure, the average concentration of PFO in whole fish was 46.7 {micro}g/g, wet wt., and did not differ significantly from the concentration measured in fish after 8 days of exposure (44.7 {micro}g/g, wet wt.). The 13 day apparent steady-state bioconcentration factor for PFO was 1.8. When transferred to clean water, the concentration of PFO in whole fish decreased to 5.9 {micro}g/g, wet wt., in 4 days and remained constant throughout the remainder of the depuration experiment.
