Temporal changes in CYP1A, DNA adducts, and contaminant residues in fish removed from contaminated sites

In combined field and laboratory studies, English sole (Parophrys vetulus) were captured from two highly polluted areas and from a relatively, uncontaminated area. The Duwamish Waterway contains a variety of contaminants, including polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs), whereas the other contaminated area, Eagle Harbor, is contaminated primarily with creosote-derived PAHs. Fish were sampled at time of capture for analyses of several biomarkers of contaminant exposure and effects, including hepatic cytochrome P4501A (CYP1A), biliary fluorescent aromatic compounds (FACs), liver, muscle, and brain PCBs, and levels of hepatic DNA adducts. Remaining fish were transported to the laboratory, maintained in clean flowing seawater, and sampled after 3, 7, 14, 21, 28, 42, and 56 days. CYP1A and biliary FACs declined steadily in fish removed from the polluted areas. Estimated half-lives ranged from 14 to 16 days for CYP1A and 12 to 28 days for biliary FACs. PCB levels in the liver tended to increase over time, suggesting that PCBs were not primarily responsible for induction of hepatic CYP1A in these fish. PCBs in the brain showed a similar increase with time, while PCBs in muscle tissue decreased. Half-lives of hepatic DNA adducts ranged from 40--100 days. There were no substantial differences in biomarker half-lives between the two polluted areas. These results are consistent with earlier laboratory studies, and assist in interpreting results of field surveys of contaminant exposure and effects in aquatic organisms. Subsequent to this study, Eagle Harbor has undergone remediation by capping, and the results of this work will enhance the assessment of the efficacy of this effort.
