Brain and cardiac uptake of I-123 IPT: An analog of cocaine

I-123 N-((E)3-Iodopropen-2-yl)-2b-Carbomethoxy-3b-(4-chlorophenyl)tropane (IPT) is an analog of cocaine that has a high affinity for dopamine reuptake sites (Kd = 0.7 nM in rat striatal membrane preparations). I-123 IPT may be a potential SPECT imaging agent for evaluating presynaptic dopaminergic integrity. The purpose of this study was to measure the biodistribution of I-123 IPT in a non-human primate and estimate its radiation dosimetry in humans. Transmission scans were performed on a 3.5 kg cynomologous monkey with a sheet source containing 150 MBq of I-123. A bolus of 185 MBq was then administered intravenously. A series of conjugate whole body scans were subsequently acquired for 20 minutes each over the next 15 hours on a dual headed camera. Regions of lifting (ROIs) were placed around 15 organs on the first conjugate pair of whole body images and transposed as a single set onto all the other scans. Decay and attenuation corrections were applied. The geometric mean activity in each ROI was compared to the activity in the net injected dose. Biexponential functions were fit to the time-activity curves for each organ and integrated to calculate die residence times.
