Low frequency of Gln{sub 353} allele of the Arg/Gln{sub 353} polymorphism of the factor VII (FVII) gene in the Chinese

Plasma factor VII activity is one of the independent risk factors for CAD. The Chinese have low incidence and mortality from CAD compared to Caucasians. An association of the Arg/Gln{sub 353} polymorphism of the FVII gene with FVII activity has been observed in several studies. We have studied the distribution of the Arg/Gln{sub 353} polymorphism of the FVII gene in 230 Chinese adults of both sexes (107 m, 123 f). A 312 bp-long region of exon 8 of the gene continuing a variable MspI site and a nearby constant MspI site was amplified by polymerase chain reaction with primers of published sequences. The presence of the cutting site represents the common Arg{sub 353} allele while the rare allele (Gln{sub 353}) has no cutting site. The frequency of Gln{sub 353} in the Chinese has been found to be low (0.04 in the males and 0.05 in the females) compared to that in the Caucasians (0.10). Genotype distribution of this polymorphism was at Hardy-Weinberg equilibrium.
