Cloning of the genomic locus on chromosome 13 of a new human homeobox gene with strong homology to the rat somatostatin transactivating factor-1

Degenerate PCR primers designed to amplify highly conserved homeobox motifs were used to amplify a chromosome 13-specific genomic phage library. A resulting PCR product was sequenced and found to have high homology to the Xenopus gene, Xhox 8. This PCR fragment was used to screen a chromosome 13-specific genomic cosmid library. Several cosmids were recovered and homeobox containing subclones were sequenced. The homeobox contained in these chromosome 13 cosmids was found by blast analysis to have near-perfect homology at the amino acid level to the rat somatostatin transactivating factor-1. The homeobox-containing cosmid clones were used as FISH probes and were found to map to the 13q12 region. Several cDNA libraries were screened with the homeobox-containing genomic clones, however, no positive colonies were recovered.
