Telomere reduction is not a generalized phenomenon of solid tissue neoplasms

The telomere is that region of DNA found at the ends of chromosomes which is required for replication, stability and prevention of illegitimate recombination. Telomeric DNA consists of terminal repeat arrays of base pairs characterized by clusters of G residues in the three prime strand (TTAGGG){sub a}. Telomere reduction (TR) has been identified in giant cell tumor of bone (GCT), colon carcinoma and senescent fibroblasts. To determine if TR is a general phenomenon of oncogenesis we examined a wide variety of human solid neoplasms and age-matched controls. Using Southern blotting techniques, a radioactive telomeric probe (TTAGGG){sub 50} was hybridized to genomic DNA digested with HinfI and signal DNA peak migration distance was determined by densitometry. In telomeric reduction the migration distance is greater than in the control while telomeric elongation shows the opposite finding.
