Identification of transcripts that are differentially expressed in normal and cancerous prostate tissue

To identify candidate genes for prostate cancer, mRNA isolated from normal prostate and from two prostate cancer cell lines was screened for differentially expressed sequences. The cell lines included LnCaP, an androgen-sensitive cell-line derived from a metastatic tumor of a 55-year old Caucasian male, and LnO, and androgen-insensitive line derived from LnCaP. mRNA from these cell lines, as well as mRNA from normal prostate tissue, was subjected to differential display. The mRNA was reverse transcribed with one of the following primers: T{sub 11}MA or T{sub 11}MG (M=A, G, or C). Reverse-transcribed sequences were PCR-amplified using the same polyT primer and a series of arbitrary ten-base oligonucleotides, with radioactive nucleotides included in the amplification reactions to permit subsequent visualization of the products on polyacrylamide gels. Several sequences have been identified that are differentially displayed in mRNA derived from normal prostate tissue compared with PCR-amplified, reverse-transcribed mRNA from the tumor cell lines. These transcripts represent candidates for tumor suppressors that are underexpressed in the prostate cancers from which the cell lines were derived. Clones of the differentially displayed sequences will be used to isolate cDNA clones for further analysis, including validation of differential expression using Northern blots and DNA sequence analysis to determine potential sequence homology with other genes known to be involved in cancer etiology. Additional differential display experiments will also be carried out to identify more candidate tumor supressor mRNAs and to identify sequences that may be expressed in tumors but not in normal prostate.
