Synthesis and initial evaluation of multidentate Me-3,2-HOPO ligands for in vivo Pu(IV) chelation

Ligands that stably bind Pu(IV) at physiological pH are needed to enhance its excretion after accidental human contamination. Based on the similarity in coordination properties between Fe(III) and Pu(IV), a new class of multidentate hydroxypyridinone (Me-3,2-HOPO) ligands has been synthesized. Three ligands have been found to be particularly effective, with low toxicity: TREN-(Me-3,2-HOPO) and two other Me-3,2-HOPO ligands. When injected or given orally, these have proven to be highly effective for chelation of Pu in vivo. In addition, TREN-(Me-3,2-HOPO) was found to be an exceptionally good in vivo chelator of Am(III); when injected, it also reduces the body content of Np(V) and reduces kidney U(VI) content effectively.
