(Genetic engineering with a gene encoding a soybean storage protein)
This report summarizes the progress on the grant. The purpose of our research is to study the expression of genes encoding ..beta..-conglycinin, a seed storage protein of soybeans. In previous reports we described the isolation and characterization of one such gene, that encoding the ..cap alpha..'-subunit of ..beta..-conglycinin. Characterization included DNA sequence analysis, and comparison with comparable genes of Phaseolus and Peseine. A reprint of the JBC paper is included with this report. A primary reason for determining the sequence and for making the comparison with the phaseolin gene sequence was to attempt to identify the DNA sequences that are responsible for regulating the expression of the ..cap alpha..'-gene. The comparison reported in the JBC paper was very helpful; however, it was only comparative and we then extended our work to functionally identify the sequences that are important for the expression of this gene.
