Title: In vitro heat shock increases the synthesis of Cu-Zn superoxide dismutase by normal mammalian tissue

The harmful consequences of numerous physiological stresses, including heat shock, may be due in part to increased cellular production of superoxide. The purpose of this study was to determine whether Cu-Zn superoxide dismutase (SOD) activity is altered during the lung's response to in vitro heat shock. Adult rat lungs were perfused free of blood, sliced and incubated in Krebs Ringer bicarbonate medium containing 0.7 mM (/sup 3/H)phenylalanine (phe) at 37/sup 0/C or 42/sup 0/C. Cu-Zn SOD activity was unchanged after 2h incubation at 42/sup 0/C, but was increased 12% (p<.05) after 4h incubation. Lung protein synthesis (nmol phe incorp/mg DNA/h) was decreased 24% (p<.05) during the initial 2h incubation at 42/sup 0/C and remained 21% lower than control for the following 2h at 42/sup 0/C (p<.05). In contrast, Cu-Zn SOD synthesis was unchanged during the initial 2h at 42/sup 0/C and was increased 86% (p<.02) during the following 2h at 42/sup 0/C. These data indicate for the first time that SOD induction occurs in response to heat shock in normal mammalian tissue. Heat shock proteins are typically induced within 5-30 minutes of the onset of hyperthermia, hence the present results suggest that Cu-Zn SOD induction is not part of the lung's initial response to heat shock, but may be secondary to an increase in cellular superoxide production.
