Refining the chromosome 13 linkage map

Employing the CHLC (Cooperative Human Linkage Center) skeletal map and likely location tables for chromosome 13 (based primarily on CEPH families) as a starting point for the choice of several markers, we have typed 10 very polymorphic markers on two highly consanguineous (non-CEPH) kindreds. 98 samples were typed in these kindreds which contain a total of 154 individuals. Linkage and haplotype reconstruction indicate a clustering of (pter-..D13S115-5%-D13S232-10%-D13S120-5%-D13S192). This confirms the order and approximate distances given by CHLC for the first three markers and locates D13S192 (not on the CHLC maps) distal to them on proximal 13q12. Tight linkage of GGAT3D08 and GATA6B07 is confirmed with a peak lod score >2 at 0% recombination. This cluster is >10 cM distal to the D13S115 - D13S192 cluster based on no positive lod scores for any pairwise between-cluster analyses and exclusion to nearly 10% recombination for some pairwise between-cluster analyses. This is consistent with the CHLC distance of at least 20 cM between D13S120 and GGAT3D08 on their map. Pairwise lod scores also suggest that the tetranucleotide marker GATA10D05, which CHLC had only assigned generically to 13 as of Dec, `93, is closely linked to D13S318 (GATA5H09) (Z>2.0 at {theta}=0.0) and that marker GATA2A02 may also be close by, although its likely location in the CHLC tables is some 30 cM from D13S318. No positive lod scores were observed between these markers and markers in the two proximal groups. Additional markers and kindreds are being typed to strengthen these independent mapping data.
