Ocular tumors in hybrid fish of the genus Xiphophorus exposed to n-methyl-n-nitrosourea (mnu)

The authors investigated the potential of hybrid fish, genus Xiphophorus, as a useful experimental animal model for human retinoblastoma, a neuroepithelial tumor. These fish have been valuable for over 60 years in studies of induction of neoplasia by chemical and physical carcinogens. There is extensive homology between the human and fish Rb genes. Some of the pigmented and non pigmented fish from the first backcross generation developed unilateral ocular tumors after treatment with MNU. The yield of growths, at 4-6 mo, among 1,200 fish increased with the product of the concentration times treatment time times number of treatments. A preliminary dose response curve was concave upwards rising from a control value of 0/185 to 15% at high doses. A few growths have been examined histologically. They were protruberant meduloepitheliomas, originating from the same stem cells that give rise to human retinoblastoma.
